API 5 is an endogenous Inhibitor of Caspase-2!

Caspases are key enzymes responsible for mediating apoptosis and initiator caspases are activated by dimerization.


Work from MSU Mainz now deciphered the first endogenous inhibitor of caspase-2. Caspase-2 was the first mammalian caspase ever cloned but despite rigorous investigations its function remains poorly understood.

Original investigations carried out by MSU Mainz and now published in EMBO Reports demonstrate that API5 directly binds to the CARD domain of Caspase-2 and prevents its dimerization. Loss of API5 sensitizes cells to caspase-2-dependent apoptotic cell death.
